Human umbilical cord mesenchymal stem cell-derived extracellular vesicles ameliorate airway inflammation in a rat model of chronic obstructive pulmonary disease (COPD)

Abstract Background Chronic obstructive pulmonary disease (COPD) is an incurable and debilitating chronic characterized by progressive airflow limitation associated with abnormal levels of tissue inflammation. Therefore, stem cell-based approaches to tackle the condition are currently a focus regenerative therapies for COPD. Extracellular vesicles (EVs) released all cell types crucially involved in paracrine, extracellular communication. Recent advances field suggest that cell-derived EVs possess therapeutic potential which comparable cells their origin. Methods In this study, we assessed anti-inflammatory effects human umbilical cord mesenchymal (hUC-MSC)-derived rat model were isolated from hUC-MSCs transmission electron microscope, western blotting, nanoparticle tracking analysis. As COPD, male Sprague-Dawley rats exposed cigarette smoke up 12 weeks, followed transplantation or application hUC-MSC-derived EVs. Lung was subjected histological analysis using haematoxylin eosin staining, Alcian blue-periodic acid-Schiff (AB-PAS) immunofluorescence staining. Gene expression lung microarray Statistical analyses performed GraphPad Prism 7 version 7.0 (GraphPad Software, USA). Student’s t test used compare between 2 groups. Results Both resulted reduction peribronchial perivascular inflammation, alveolar septal thickening mononuclear decreased number goblet cells.
